It is likely to be less than 25-26, i.e., less than the number of letters in the Latin alphabet.
I see more than 30 in autonomous use.
Among other things, it is unclear how to treat combinations of a-la ee and some problematic characters. For example, s:

It might be a spelling error for EVA s or d, but it still looks strange.

A simple substitution cypher would reproduce all the patterns, sequences, letter distributions, and (crucially) entropy, of the original language. Voynichese does not even closely match any known language with which it has been compared on those measures.
